چکیده

The ribonucleotide reductase activities in vitamin B12-sufficient and -deficient cells of Euglena gracilis were measured. We found that the cells progress into vitamin B12 deficiency the enzyme activity increases, reaching a maximum value of 20-fold in advanced deficiency. No signigicant differences in the activities were found to result as a consequence of different growth conditions. We propose that the increased activity in vitamin B12-deficient cells is due to an increase in enzyme protein.

Euglena gracilis ribonucleotide reductase: the eukaryote class II enzyme and the possible antiquity of eukaryote B12 dependence.

Ribonucleotide reductases provide the building blocks for DNA synthesis. Three classes of enzymes are known, differing widely in amino acid sequence but with similar structural motives and allosteric regulation. Class I occurs in eukaryotes and aerobic prokaryotes, class II occurs in aerobic and anaerobic prokaryotes, and class III occurs in anaerobic prokaryotes. The eukaryote Euglena gracilis...

Vitamin B12 in human blood and serum. I. Comparison of microbiologic assays using normal subjects.

HE MICROORGANISMS used for Vitamin B12 (cyanocobalamin) assay are LaCtobaciUus lewhmanniz (1), Euglena gracilis (2), Ochromonas maihamensis (3-5), and a mutant strain of Escherichia COli (E. coli 113-3) (6). Improvements in protozoan assays for B12 show such assays to be more sensitive and specific than bacterial assays (2, 7, 8). Ochromonas maihamensis is highly specific for true forms of B12 ...

Identification of NADPH-thioredoxin reductase system in Euglena gracillis.

Euglena gracilis contains a protein system which can utilize the reducing power of NADPH in the ribonucleotide reductase-catalyzed reduction of CTP. The proteins required for this reaction are a flavoprotien with a molecular weight of approximately 185,000 which is functionally similar to thioredoxin reductase (NADPH), EC 1.6.4.5, and another protein (Protein I) whose function in the reaction i...

The role of vitamin B12 in the metabolism of Euglena gracilis var. bacillaris.

1. The concentrations of RNA, DNA and protein are decreased in cells of Euglena gracilis var. bacillaris grown on suboptimum concentrations of vitamin B(12). 2. The addition of vitamin B(12) to deficient cells stimulates the incorporation of [(14)C]formate into the above cell components as well as into thymine of DNA and serine and methionine of protein. 3. In a cell-free system from vitamin B(...

VITAMIN B 12 AND THE MACROMOLECULAR COMPOSITION OF EUGLENA II. Recovery from Unbalanced Growth Induced by Vitamin B 12 Deficiency

When vitamin B12 is added to B12-deficient cultures of Euglena gracilis, the cells undergo two relatively synchronous cell divisions within a shorter than usual period of time, apparently as a result of a transitory shortening of the cell cycle . The first cell division pulse, occurring 4 .5 h after addition of B12, is preceded by the completion of DNA duplication, but appears to involve no net...
